Effect of Extra Column Volume on Your Results
Below are two chromatogram examples of how two different instruments can produce very different results using the exact same conditions. They were both tested, using the exact same certified, HPLC Column, with the exact same NIST traceable standards on the exact same day. These differing results of an older and newer HPLC instrument may be simply they are plumbed quite differently.
Recommendation: Make sure all fittings are new, secure and that all tubing is well cut and as short as possible. Not all HPLC instruments offer optimal extra volume; check with your instrument manufacturer on how to optimize your system.
