These HPLC columns are made with 10um, 125A, irregular shaped silica similar to the original µBondapak® Amino column used in many USP methods today. The MicroBee™ Amino columns are held to very tight specifications for reproducibility batch to batch and have a 3.5% carbon loading.
Amino columns are used in normal phase HPLC mode most often and can also be used in HILIC mode.
The amino functionality of the amino-propyl ligand has a lone electron pair. Therefore, the hydrocarbon chain (C3) will be polar and will have a slightly negative dipole moment toward the Nitrogen of the amino group. This makes the stationary phase more polar than plain silica.
Different molecules will have different retention based on their polarity and their size (access to the ligand) thus, a unique Selectivity.
